چکیده
The effects of nitrogen mustard, Myleran, and x-radiation on the incorporation of isotopic orotic acid, aspartic acid, formaldehyde, adenine, and phosphate into the nucleic acids of the L1210 ascites leukemia are reported. The lack of uniformity in the incorporation pattern following pretreatment with each of the three drugs was considered significant. The treatments could result in stimulation as well as inhibition of precursor utilization. I t was concluded that the alkylating agents attack numerous sites with varying degrees of specificity.

On the mechanism of action of the alkylating agents. I. Interaction of alkylating agents with nucleic acids.
The incorporation of isotopic ethylene-C1Mmino-s-triazine (TEM), Myleran, chlorambucil, and sulfur mustard into the DNA fraction of regenerating liver and of various mouse ascites lymphomas was measured. The in vivo incorporation of these alkylating agents was estimated to be of the order of 1 mole of drug per mole of DNA polymer. Results obtained by several different methods of isolation of DN...

In the field of clinical cancer chemotherapy the alkylating agents are second only to androgens and estrogens in frequency of use. Since the first description of the effects of nitrogen mustard in problems of human neoplastic disease, congeners have been synthesized and tested in the laboratory and clinic. The objectives of this developmental program have been to increase anti-tumor specificity...

Young adult female RF mice were subjected to four successive midlethal doses of nitrogen mustard (HN2), triethylene melamine (TEM), 1 ,4-dimethanesulfonoxy butane (myleran), or whole-body x-rays. The agents were given at 14-day intervals, and the survivors were observed for effects on longevity and on the incidence of late occurring diseases. The percentages of mice dying within 30 days after t...
متن کاملThe reaction of mustard gas with nucleic acids in vitro and in vivo.
Mustard gas [di-(2-chloroethyl) sulphide] and analogous alkylating agents produce a wide range of biological effects (cf. review by Boyland, 1948). Some ofthese effects, such as induction ofmutations (Auerbach & Robson, 1946) and interference with mitosis and chromosome breakage (Darlington & Koller, 1947), stimulated interest in the reactions of alkylating agents with deoxyribonucleic acid. Pr...
متن کاملEffects of uracil mustard on DNA, RNA, and protein biosynthesis in tissues of A-J mice.
The effects of 5-bis(2-chloroethyl)aminouracil (uracil mustard) on some of the biochemical events occurring in lung were studied since this compound, a reactive alkylating agent and analog of uracil, markedly increased the incidence and number of lung tumors in strain A/J mice. Similar measurements were per formed in kidney and liver, organs which do not undergo a tumorigenic res]x>nse to uraci...
